Depolt was the son of Jan I, Freiherr Popel z Lobkowicz, and Anna Svihovska z Ryzmberka.
During the struggles for power between Georg Podiebrad's followers, including Johann II z Rozmberk, and the 'Grünberg Alliance' of nobles opposed to Georg, their leader Zdenek ze Sternberka, an arch-enemy of Rozmberk, captured Rozmberk nad Vltavou in January 1469, arrested Depolt and his father and held them in his castle of Krumau (Cesky Krumlov). Jan Popel fell ill in captivity and died in 1470. Depolt continued to be held in captivity until 1475.
In 1502 Depolt acquired the estate of Bilin (Bilina) north of Prague from the Colditz family, and in 1527 he added the estate of Dux (Duchcov) to it.
Depolt was married twice, first to Johana ze Svamberka, daughter of Bohuslaw ze Svamberka and his first wife Ludmilla von Rosenberg. Johana, recorded alive in 1490, probably died in 1491. There was no progeny from this marriage. Depolt's second marriage was to Anna Micanova Klinstejna, daughter of Litvin Klinstejna and Majdalena z Vrtby. There were nine children from his second marriage, of whom Jiri, Litvin, Christof Popel, Václav and Jan IV 'Mladsi' would have progeny, though Depolt's line, known as the Bilin line, would continue through Václav.
Depolt died at Bilin on 3 April 1527. His son Václav inherited Bilin.
