Claude was the daughter of Jean II de Brosse, seigneur de Boussac, de Sainte-Severe, and Nicole de Châtillon de Blois, comtesse de Penthièvre, vicomtesse de Limoges.
On 11 November 1485 she became the second wife of Philippe I, duke of Savoy, comte de Bresse, son of Louis, duke of Savoy, and Anne de Lusignan. They had six children of whom Charles III and Philippe would have progeny.
Although most sources identify Claude's brother Jean III as the head of the Penthièvre family in 1488 at the death of François II as the last male duke of Brittany, some sources indicate that Claude had succeeded as countess of Penthièvre on the death of her father in 1483, by right of her mother. In any event it was Jean who asserted the family's claim to the duchy of Brittany, though François' daughter Anne succeeded, and went on to marry Charles VIII, king of France in 1491.
Claude died on 13 October 1513.
